摘  要:目的研究c-myc反义寡核苷酸(ASODN)对人恶性黑素瘤细胞c-myc mRNA表达及c-myc蛋白水平的影响。方法设空白和混杂链(SODN)对照,人工合成c-myc ASODN,以30μmol/L ASODN转染人恶性黑素瘤细胞株A375,分别用原位杂交、免疫组化SABC法检测c-myc mRNA的表达及其蛋白水平。结果转染24 h后,ASODN组细胞c-myc mRNA较空白对照组、SODN组均显著下调(P均<0.01);转染48 h后,与空白对照组、SODN组相比,ASODN组细胞的c-myc蛋白水平均显著下降(P均<0.01)。结论c-myc ASODN可下调A375细胞c-myc mRNA的表达和c-myc蛋白水平。Objective To study the effects of c-myc antisense oligodexynucleotide (ASODN) on the expression of c-myc mRNA and its protein in melanoma cells. Methods c-myc ASODN was synthesized and transfected into melanoma cell line A375. c-myc mRNA and protein were detected by in situ hybridization and immunohistochemistry, respectively. Results At 24h after incubation, the expression of c-myc mRNA was significantly lower in the ASODN group than that in SODN group as well as that in the control group( P 〈 0.01 ) ;compared to those of empty control and SODN group, the level of c-myc protein in the ASODN group significantly decreased at 48h after incubation( P 〈 0.01 ). Conclusion c-myc ASODN can down - regulate the expression of c-myc mRNA and its protein in A375 cells.
