摘  要:目的在体外实验中研究脂膜微囊承载抗胶质瘤药物紫杉醇抑制C6胶质瘤细胞的作用,并探讨脂膜微囊靶向性抗瘤的机理。方法应用紫杉醇-脂膜微囊(Taxol-LCM)或单用脂膜微囊(LCM)处理C6细胞系,后用免疫荧光显微镜或激光共聚焦显微镜观察脂膜微囊在肿瘤细胞内的分布特点,用药前后肿瘤细胞形态的改变和细胞内超微结构的变化,以及动态的观察脂膜微囊进入细胞的全过程。结果紫杉醇(Taxol)可结合于脂膜微囊(LCM),可以被胶质瘤细胞内吞,起到很强的的杀伤肿瘤细胞的作用;其内吞过程为脂膜微囊先结合于细胞膜表面,后在细胞内重新分布,最终被细胞浆内的酸性成分所降解。结论脂膜微囊可以承载紫杉醇在体外起到杀伤胶质瘤细胞的作用,且脂膜微囊的代谢与细胞内的酸性成分有关。Objective To investigate the inhibation of C6 glioma cells by Taxol carried by lipidcoated microbubbles (LCM), and to discuss the mechanisms of lipid-coated microbubbles targeting tumor cells. Methods C6 glioma cell line's were treated by Taxol-LCM or LCM. The distribution of LCM, and the changes of cell morphology and ultrastructure were observed by immunoflurescent microscopy and confocal microscopy. The process of LCMs entering the glioma cells were observed dynamicly. Results Taxol may connected with lipid-coated microbubbles and the compound was endocyted by C6 glioma and acted killing effect. The whole endocyte process was that lipid-coated microbubbles first connected with the surface of glioma cells, and then redistributed in cells. Finally they were degraded in the cells associated with the acidic component of cytoplasm. Conclusions Lipid-coated microbubbles may carry Taxol to C6 glioma cells, and the compound may act an killing effect in vitro. The mechanisms of metabolism associated with acidic component.
