摘  要:目的探讨bFGF、ATRA、BMP-2对神经干细胞分化的诱导作用及对RARαmRNA表达的影响。方法采用分别含bFGF、ATRA、BMP-2及其不同组合配方的培养液培养神经干细胞,通过免疫荧光染色观测细胞分化情况,应用RT-PCR法检测其对RARαmRNA表达影响。结果bFGF无明显诱导神经干细胞分化作用,但能增加分化后细胞的存活能力;神经干细胞能被ATRA诱导成为神经元样细胞,被BMP-2诱导成为星形胶质细胞,且在不同组合配方培养液中,诱导效应不同;ATRA或BMP-2在bFGF存在的情况下,能促进神经干细胞RARαmRNA的表达。结论bFGF本身不诱导RARαmRNA的表达,但能增强神经干细胞分化后的存活并影响ATRA、BMP-2对RARαmRNA表达的诱导;ATRA、BMP-2对神经干细胞的分化具诱导功能,但二者可能具有负协同作用。Objective To determine the effects of bFGF, BMP-2 and ATRA on the differentiation and RARα mRNA expression of neural stem cells(NSCs). Methods NSCs was cultured with the mediums containing bFGF, BMP-2 or ATRA respectively. Their effects of inducing differentiation on NSCs were observed by immunofluorescence staining and RARer mRNA expressions of NSCs were determined by RTPCR. Results bFGF couldn t induce NSCs to differentiate. But, it was very important to the survival of cells in the process of differentiatiion. NSCs were induced to differentiate into neuron-like cells by ATRA while into astrocytes by BMP-2; and the effects were different with medium containing inducing factors of different formula. BMP-2 or ATRA could increase the expression of RARα mRNA with the medium containing bFGF. Conclusions The bFGF can't enhance the expression of RARer mRNA alone, but it can affect the process of expression of RARαmRNA induced by ATRA or BMP-2. ATRA and BMP-2 can induce the differentiation of NSCs. ATRA and BMP-2 antagonize each other in their effects on NSCs differentiation.
