摘 要:目的观察谷氨酰胺对半乳糖胺/脂多糖(Galn/LPS)诱导急性肝损伤发生的影响。方法采用Galn/LPS所致急性肝损伤模型,雄性昆明种小鼠在注射Galn/LPS(7mg/4μg/只)前30min,自腹腔注射3.5%的谷氨酰胺溶液(1g/kg体重),分别于注射Galn/LPS6h后测定血浆谷丙转氨酶(ALT)活性、肝脏组织丙二醛(MDA)、谷胱甘肽(GSH)、肿瘤坏死因子α(TNFα)的含量。结果谷氨酰胺预处理可抑制Galn/LPS导致的血浆ALT活性、肝组织TNFα、MDA水平的升高,维持GSH的浓度。结论谷氨酰胺维持肝脏GSH含量,增强抗氧化应激的能力,抑制TNFα的分泌,从而减轻Galn/LPS引起的急性肝损伤。Objective To investigate the effect of glutamine on acute liver injury by Galn/LPS. Methods The acute liver injury model was established by Galn/LPS. Pretreatment with 3.5 % glutamine solution by intraperitoneal at the dose of 1 g/kg weight. Male mice were treated with LPS plus D-galactosamine. Then blood removed from the vena was analyzed for ALT, and liver tissue was analyzed for the density of MDA, GSH and TNF-α. Results Pretreatment with glutamine could abrogate the high levels of plasma ALT activity, the density of TNF-α and MDA in liver by Galn/LPS, and maintain the density of GSH. Conclusion Glutarnine can maintain the level of GSH in liver tissue,regulate the redox state,and prevent liver injury induced by Galn/LPS.
