摘 要:本文应用组织培养、免疫细胞化学和电生理方法研究了体外培养条件下小脑髓鞘形成及电生理表现。结果表明,体外培养13d时,电镜下即可见到轴突周围有少量髓鞘板层。培养14d时,光镜下观察到,髓鞘为两条平行双折光结构,主要分布在髓质;在皮质和核区有髓鞘碱性蛋白阳性的少突胶质细胞集中分布。体外培养24h,在小脑皮质可记录到频率约为80~110次/s,幅值0.2mV的自发放电;至9div时,可记录到较大的自发放电,频率约为40~60次/s,幅值0.6mV。本文对体外培养条件下中枢神经系统髓鞘形成及小脑发育各阶段电生理表现进行了讨论。The morphological observations of myelination and its electrophysiological recordings in newborn B6C3 mouse cerebellum in vitro were studied by means of the techniques of tissue culture,immunocytochemsitry and electrophysiology,The myelin sheaths could be observed under TEM on 13 days in vitro(div)and could also be seen under LM on 14 div in characteristics of two parallel lines and double-dioptric maner,most of which located in the medulla. The myelin basic protein(MBP)positive oligodendrocytes concentrated in the cortex and deep nuclear area. The spontaneous discharges of the cerebellum explants were also recorded.Their frquency and amplitude were 80-110/s and 0.2 mV respectively at 24 hours in vitro,and the larger spontaneous discharges with a frquency and amplitude about 40-60/s and 0.6 mV respectively were recorded on the 9 div,The CNS myelination and the electrophysiological properties of cerebellum in different developmental stages in vitro were discussed.
