摘  要:目的联合检测细胞角蛋白20(cytoker-atin20,CK20)表达和端粒酶的活性,初步探讨它们在检测大肠癌微转移中的意义。方法用RT-PCR法,检测52例大肠癌患者术前和术后不同时间外周血中的CK20的表达。用端粒重复片段扩增方法检测相应患者外周血中的端粒酶的活性。结果52例患者外周血中检出CK20表达及端粒酶活性检测阳性率按不同时间段分别为61·9%与57·7%(术前),69·1%与72·1%(术后24h),34·6%与30·9%(术后早期化疗结束后)。两者的阳性表达均与患者Duke’s分期存在显著性相关。结论端粒酶和CK20均可作为标志物来检测大肠癌患者外周血中微转移。OBJECTIVE: To detect cytokeratin 20(CK20) mRNA, telomerase activity and their correlation with micrometastasis of colorectal carcinoma. METHODS: CK20 mRNA and telomerase activity in peripheral blood of 52 patients with colorectal carcinomas were detected at different time points before and after operation by RT-PCR and TRAP respectively. RESULTS: At different phases, the positive rates of CK20 mRNA and telomerase activity of those patients were 61.9% vs 57.7 (before operation), 69.1% vs 72.1% (24 h after operation) and 34.6% vs 30.9% (after the short-term chemotherapy) respectively. CONCLUSION: CK20 mRNA and telomerase activity may be potential tumor markers for detecting micrometastasis and recurrence in peripheral blood.
