摘  要:目的:观察力竭游泳对红细胞膜脂质过氧化反应的作用以及对红细胞变形性的影响。方法:实验于2005-04在中国医科大学生化实验室完成。选用普通级SD雄性大鼠24只,随机分为4组:对照组,运动后即刻组,运动后lh组,运动后24h组,每组6只。运动组大鼠在水温32℃的玻璃泳槽中做无负重的力竭性游泳。对照组不做游泳。力竭标准以大鼠下沉后10s不露出水面为标准,迅速取出大鼠擦干,按不同时期运动后即刻,运动后1h,24h对大鼠进行摘眼球取血,肝素抗凝。制备红细胞膜,红细胞膜蛋白含量的测定采用Bradford蛋白定量法。红细胞膜丙二醛含量采用荧光法测定,红细胞变形性的测定采用Judlklewlcy改进的离心群集法。评估标准:红细胞群集差越大,其变形性越大。通过t检验作统计学处理。结果:在整个实验过程中无大鼠死亡,全部进入结果分析。①力竭游泳运动后即刻组和运动后1h组、运动后24h组红细胞膜上的脂质过氧化产物丙二醛显著高于对照组(0.629±0.087),(0.593±0.062),(0.542±0.079),(0.476±0.083)μmol/g;P<0.01。大鼠力竭游泳运动后即刻组和运动后1h组红细胞群集差与对照组相比明显增高(14.51±11.16),(14.90±1.12),(12.80±1.66);P<0.01,即红细胞变形性在运动后即刻和1h后极显著下降,运动后24h组红细胞群集差基本恢复正常,说明红细胞变形性基本恢复正常。②红细胞群集差与红细胞膜上丙二醛呈显著正相关,相关系数为0.517(P<0.05),即红细胞变形性与红细胞膜上丙二醛含量呈负相关(P<0.05)。结论:力竭游泳能够引起大鼠红细胞脂质过氧化反应发生,可造成对红细胞的直接损伤,从而导致红细胞变形性下降。红细胞变形性下降很有可能是运动性贫血的重要诱因。有关力竭游泳对血液流变学影响方面的深入研究,将有助于阐明运动对红细胞的损伤以及运动性贫血发生的机制。AIM: To observe the influence of exhaustive swimming on the lipid peroxidation of erythrocytic membrane and erythroeytic deformability. METHODS: The experiment was carried out in the biochemical lab of China Medical University in April 2005. Twenty-four male SD rats of common degree were randomly divided into 4 groups with 6 rats in each group: control group, immediately, 1-hour and 24-hour postexercise groups. Rats in the exercise groups swam in the glass made swimming chamber of 32℃ without loading until exhaustion, and those in the control group did not swim. The standard of exhaustion was that the rats could not emerge from the surface of water after submerge for 10 s, and then the rats were rapidly taken out and dried, the eyeballs were extracted and blood was drawn at different points (immediately, 1 and 24 hours) after exercise, and then anti-coagulated with heparin. Erythrocytic membrane was prepared, and the content of erythrocytic membrane protein was detected with Bradford protein quantitative method, the content of erythrocytic membrane malondialdehyde was determined with the fluorescent method, the erythrocytic deformability was detected with Judlklewlcy modified centrifugated clustering method. Evaluation criteria: the greater the difference of erythrocytic cluster, the greater the deformability. The t test was applied in the statistical analysis. RESULTS: No rat died during the experiment, and all were involved in the analysis of results. ①The content of malondialdehyde (product of lipid peroxidation on erythrocytic membrane) was significantly higher in the immediately, 1-hour and 24-hour postexercise groups than in the control group [(0.629±0.087), (0.593±0.062), (0.542±0.079), (0.476±0.083) μmol/g, P 〈 0.01]. The differences of erythrocytic cluster in the immediately and 1- hour postexercise groups were obviously increased as compared with that in the control group [(14.51±11.16), (14.90±1.12), (12.80±1.66), P 〈 0.01], it was indieated
