摘  要:目的探讨NFκB核酸对增殖的血管平滑肌细胞炎症反应和凋亡的影响。方法大鼠胸主动脉平滑肌细胞传代培养,实验共分六组。阳离子脂质体转染法将NFκB寡核苷酸导入血管平滑肌细胞,检测细胞凋亡、细胞内NFκBp65基因表达以及ICAM1、VCAM1、MCP1蛋白合成情况。结果血管平滑肌细胞转染NFκB反义或诱骗寡核苷酸后,细胞凋亡增强,凋亡时间延长。反义组NFκBp65蛋白质表达较正义组降低(P<0.05);反义+诱骗联合干预并未优于反义组单独治疗,但与诱骗组相比NFκBp65的蛋白质表达降低(P<0.05)。反义组、诱骗组的ICAM1、VCAM1、MCP1蛋白质表达较相应的对照正义组、诱骗对照组均降低(P<0.05)。结论NFκB的反义和诱骗寡核苷酸均能抑制血管平滑肌细胞ICAM1、VCAM1、MCP1的蛋白质表达,抑制血管平滑肌细胞增殖,使细胞凋亡增强;NFκB反义寡核苷酸及反义+诱骗寡核苷酸联合干预可减少NFκB生成。Objective To investigate effect of nuclear factor-kB(NF-kB) on inflammation and apoptosis of vascular smooth muscle cells(VSMCs). Methods Rat aortic smooth muscle cells were divided into 6 groups randomly. Oligonucleotide of NF-kB were transfered into VSMCs by means of lipofectamine-oligonucleotides compounds. Apoptosis of VSMCs was detected as well as local gene expression and protein synthesis of NF-kB and ICAM-1, VCAM-1 and MCP-1 protein synthesis in VSMCs. Results VSMCs apoptosis reinforced and apoptosis time elongated. NF-kBp65 protein synthesis in antisense group decreased 29.58% compared with sense group( P〈0.05). There was no significant difference statistically between decoy group and scramble group. Antisense plus decoy group was not superior to that of antisense group, but combination of decoy and antisense oligonucleotide lowered NF-kB p65 protein synthesis more obviously than decoy group. ICAM-1 , VCAM-1 and MCP-1 protein synthesis in antisense group and decoy group decreased, compared with sense group and scramble group, respectively. Conclusion Decoy and antisense oligonucleotide of NF-kB could obviously inhibit ICAM-1, VCAM-1 and MCP-1 protein synthesis in VSMCs. Decoy and antisense oligonucleotide of NF-kB might inhibit proliferation of VSMCs induced by TNF-α and reinforced cellular apoptosis. Production of NF-kB decreases under antisense group and combination of antisense and decoy oligonucleotide of NF-kB.
