摘 要:本文对迄今通用的平皿琼脂糖孔穴扩散法进行了改进,使检测敏感性提高2.2~3.8倍,抗菌肽含量为0.1μg时即可观察到有明显的抑菌圈,薄层平板法不仅可保证琼脂糖厚度均匀一致,从而提高检测结果的稳定性,而且可由原来的半定量发展为定量测定。此法可广泛应用农业、医药,对各种抗菌物质作微量测定及对不同抗菌物质的抑菌活性进行比较。The commonly used agarose diffusion assay on glass-plate was modified by using a thin-lay-er agarose plate for detection of biological activity of antibacterial polypeptide. By using this method,thesensitivity was increased 2.2~3.8 times.When 0.1 μg antibacterial polypeptide,Shiva l.was loaded to thehole on agarose thin-layer plate,the antibacterial transparent zone could be cleary defined.This method in-sures an even thickness of agarose plate so that the results are reproducible. While previous agarosedif-fusion method on glass-plate can only detect semi-quantitatively,thes method provides a quantitative assayof antibacterial substances.The method is believed to be able to widely used in agricultural and medicalfields for assaying antibacterial compounds and for comparing the biological activity of different antibacteri-al substances.
