摘 要:利用KCN、H_2O2和SDS的选择性反应引起的SOD同工酶谱带变化即可鉴别粗抽提液中的SOD同工酶类型;用这种方法对五株不同种根霉的鉴别实验表明,它们均不同程度地含有Cu,Zn型和Mn型两种SOD,前者约占80%左右,后者只占20%左右。用邻苯三酚自氧化法对样品中SOD活性测定结果与在同工酶谱上的鉴别结果基本一致。This paper is dealing with an identification of SOD isozyme types form Rhizopus crude extracts on the PAGE gels in the presence of KCN,H2O2 and SDS pretreatment. The results from both isozymo grams and 1-D laser scanning patterns showed that all of the strains tested, F1(R. spp.), AS 3. 866 (R.oryzae Went et Prinsen-Geerligs),AS 3. 2667 (R. arrhizus Fisher),Q303(R. spp.) and AS 3. 1136 (R. chinensis Saito), contained both Cu, Zn type and Mn type SOD at the same extent,i. e. the Cu,Zn-SOD up to about 80%,and the Mn-SOD only about 20%.In oder to verify the identification,SOD activities of the samples were also assayed before and after being treated with the three chemical agents. Both results were coincided.
