摘  要:它莫昔芬(Tamoxifen)是临床乳腺癌内分泌治疗中重要的抗雌激素药物之一,长期用药可产生耐药性。为了阐明耐药性发生的机理。本文作者应用改进的Hugs双层琼脂培养法,对SHZ-88细胞进行克隆培养,建立了耐受它莫昔芬的大鼠乳腺癌细胞株y2。该细胞株对药物它莫昔芬的敏感性比原代细胞降低了10倍,但对雌激素的敏感性没有变化。本细胞株的建立将有助于研究耐它莫昔芬乳腺肿瘤细胞的生物学特性以及作为寻找和筛选抗耐药肿瘤药物的模型。Tamoxifen is one of the important antiestroge nic hormones in pharmacotherapy for endocrine-fcsponsive human breast cancer , but the therape utic effect is gradually decreasing due to drug-resistance of tumor Inthe l on g- term adm inistration. In order to fi nd out the ta moxifen- resistant mechan ism,the authors established anexperim en tal mold el in vivo andinvitro by improved Hug’ s double- layer soft agar culture. A tamoxifen-resistan t ratmammary tumorcel l strain(de signat ed as y2 cell)was isolated by clonal culture from SHZ-88 cell line. The y2 cell istentimesinsensitivetotamoxifenthanParentcellline,buthasnochangeofsensitivitytoestrogens. The authorssuggest that establishment of y2 cell strain can proide a practical tool tostudy the biological characteristics of resistantmammarytumorcells and select the effective anticancer drug for the tamoxifen- resistant mam m ary tumors.
