Study of Potassium Cyanide Inhibiting Alanine Transaminase Activity in Isolated Damaged Hepatocytes in Fasting Rats

摘  要:使用80μmol/L的FCCP和2mmol/L的KCN制备离体大鼠肝细胞损伤模型,并观察孵育60分钟时LDH和ALT的逸出率。发现KCN明显地抑制了ALT总活性达40%以上,游离ALT活性也被显著抑制,而LDH活性无被抑制现象。KCN组ALT逸出率为68.3±3.3%与FCCP(83.2±3.9%)相比有极显著差异(P<0.001),与FCCP组ALT逸出率(82.2±2.2%)相比亦有极显著意义(P<0.001)。FCCP组的LDH逸出率为80.5±5.4%。结果表明KCN能显著抑制ALT活性。提示在用KCN制备肝细胞损伤模型时,ALT逸出率不适用于评价细胞损伤程度,此时用LDH逸出率可准确地反映细胞损伤程度。The isolated hepatocellular damage models in fasting rats were prepared by using 80μmol/L of carbonyl cyanide p-trifluoro-methoxyphenylhydrazone (FCCP) or2 mmol/L of potassium cyanide (KCN)iand the leakages (%) of lactate dehydrogenase (LDH) and alanine transaminase (ALT) were observed at end of 60-minute incubation- It was found that KCN significantly inhibited total ALT activity more than 40% and free ALT level was much more inhibited.but both of total LDH and free LDH were not affected. ALT leakage (68.3 ± 3. 3%)was significantly differed from LDH leakage,83. 2 ±3. 9%(P<0. OODin KCN group and from ALT leakage,82. 2 ± 2. 2%(P<0. 001)in FCCP group. The LDH leakage in FCCP group was 80. 5 ± 5.4% ,The results of experiments indi-cated that KCN was markedly able to inhibit ALT activity. ALT leakage is not suitble for evaluat-ing degree of cell damage when KCN is used for preparing hepatocellular damage models. It is sug-gested that LDH leakage accurately show the condition of cell damage in this situation.
