摘 要:用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)方法分析了青海高原61个春小麦品种(包括地方品种、育成品种和引进品种)的高分子量麦谷蛋白亚基。研究涉及等位基因Glu-A_1编码1、2、N(缺失)三种亚基;Glu-B_1编码7+8、7+9、6+8、20、22、17+18六种亚基;Glu-D_1,编码2+12和5+10两种亚基。同时发现带有优质亚基的品种比率低,且没有发现具有理想优质亚基组成的品种。目前种植的品种都缺优质亚基5+10,有的甚至缺亚基1或2。Giu-1位点3个等位基因的优质HMW-GS在青海高原春小麦品种中都存在。High- molecular-weight glutenin subunit (HMW-GS )COMPOSITIONS OF 61 spring wheat varieties(included 37 local varieties, 2 introduced varieties and 22 bred varieties) on the Qinghai Plateau were determined by Sodium dodecyl sulphate polyacrylamide gel electrophoresis ( SDS-pAGE).It was found that Glu-A_1 controlled 3 subunits of 1,2 and N(null); Glu-B_1 controlled 6 subunits of 7+8,7+9,6+8, 20, 22 and 17+18; Glu-D_1 controlled 2 subunits of 2+12 and 5+1.Subunit N,7+8 and 2+12 were in the majority at Glu-A_1 , Glu-B_1 and Glu-D_1,respectively.It was al-so found that varieties with good quality subunits took a lower proportion. There were no varieties with perfect quality subunit composition. All the present varieties did not have subunit 5+10,and some varieties even lack subunit 1 or2, This was the main reason for poor processing quality of spring wheat on the Qinghai Plateau. On the other hand,it was found that some varieties had good quality subunits.New varieties with perfect quality subunit composition could be bred out so that wheatquality on the plateau could be improved greatly.
