摘  要:用半消化法和组织块法接种南方鲶晚期胚,经40多代连续培养,建立了SME-Ⅰ鲶鱼细胞系。该细胞系为贴附性上皮样细胞。细胞内微丝束丰富,平行排列伸到伪足,但质膜下不很密集。细胞最适在含15%~20%小平血清的1640培养基中生长,适温28~29℃,pH6.5~7.2,群体倍增时间为58.6h,已增殖到10 ̄8个的水平。细胞染色体数为正常二倍体2n=58,但有不少异倍化细胞。该细胞系已可用于鱼类生物工程研究和进入细胞资源库保存。The paper deals with the establishment of a new fish cell line designed SME-1,which originates from the culture of late embronic cells of soutbern catfish(Silurus merid-ionalis Chen). Some important biological characteristics of the line are described briefly. The explants used for primary cuIture are tissue and semidigestive tissue. The optimalconditions for primary cuIttire and subculture are RPMI 1640 medium supplernented withmoderate amount of antibiotics, 20%new born calf serum and incubated at temperature 28~29℃,pH 6.5~7.2. A new fish cell line thus has been established through much morethan 40 subcuIture generations over a period of 3 years. The cell line appears attachment- dependent property. After attachment and expansion,it looks epithelioid cells,and itsgrowth relies on serum tightly. Microfilamentous bundles in cells dispaly long fibresthrough the center of the cells, but no thick fibres are detected under the membrane. Thegrowth ability of the line is good, and adapted to cuIture conditions strongly. The meanpopulation doubling time is 58.6 hours, and mitotic index reached to 32.66%in 48 hours. Modal chromosome number of the SME-1 cells is 58(2n=58),which takes percentageof 69.6%among total metophase,The karyotype is 2n=20 m+20 sm+14 st+4 t,NF=98.
