摘 要:本文应用小鼠神经母细胞瘤细胞A_2(NB-A_2)无血清培养建立的神经细胞老化实验研究模型,以显微荧光分光光度术测定单个细胞内脂褐素荧光值作为细胞老化指标,观察了脑啡肽(LE)对NBA_2细胞内脂褐素荧光值的影响。结果表明LE可显著降低细胞内的脂褐素荧光值(P<0.01)。提示LE可延缓NBA_2细胞的老化进程。Serum-free cultures of mouse neuroblastoma A2 (NBA2) cells were used as the experimental model for the study of neuronal aging, with microfluorometry of the autofluorescencc of lipofuscin as the index of neuronal aging. The results showed that fluorescent value (F.V.) of lipofuscin increased progressively with prolonging of culture days. The results implied that NBA2 cells in scrum-free culture change from division and reproduction into differentiation and maturation, reflecting the process of neuronal aging.After addition of Icucinc cnkephalin (LE) separately into the culture medium, the results showed that F. V. of lipofuscin was decreased in 5,10 and 15 day treatment group (P<0.01). The result suggested that LE had the property to retard the accumulation of lipofuscin and produced effect in the direction of delaying NBA2 cells iging.
