苏云金芽孢杆菌毒素对羊捻转血矛线虫第三期幼虫毒力研究

摘　　要：从人工感染捻转血矛线虫（Ｈａｅｍｏｎｃｈｕｓｃｏｎｔｏｒｔｕｓ）山羊的直肠内取出粪便，在２５℃温箱中培养７ｄ，用贝尔曼法分离出第三期幼虫，将它们加入含苏云金芽抱杆菌以色列亚种（Ｂａｃｉｌｌｕｓｔｈｕｒｉｎｇｉｅｎｓｉｓｓｕｂｓｐ．ｉｓｒａｅｌｅｎｓｉｓ）伴孢晶体毒素的培养液中，置２５℃下培养。当伴孢晶体蛋白含量为４００ｇ／ｍｌ时，幼虫的死亡率２４ｈ后为９４．７％，４８ｈ后为９７．３％。毒素对幼虫作用２４ｈ与４８ｈ，其毒力无显著差异（Ｐ＞０．０５），并且作用２４Ｌ与４８ｈ，其ＬＤ＿（５０）值伴孢晶体蛋白含量分别为８４。０５μｇ／ｍｌ和５８．６４μｇ／ｍｌ。由于伴孢晶体蛋白质对人畜无毒，可将苏云金芽孢杆菌的件孢晶体作为防治某些动物奇生虫的杀虫剂。The third-stage larvae of the ruminant parasite Haemonchus contortus were recoveredin a Baermann apparatus maintained in PBS from rectal feces of goat after incubationat 25℃ for 7 days. A toxin from the bacterium Bacillus thuringiensis(B. t.)was lethalto nematode third-stage larvae.Exposure of the third-stage larvae of the rulninantnematode Haemonchus contortus to the toxin from the B. t.at 25℃ in vitro.Treatmentof third-stage larvae of Haemonchus contortus with 400μg total protein/ml of B. t.toxinyieled 94.7%and 97.3% mortality after 24hr and 48hr respectively.It was no signifi-cant(P>0. 05) for larvicidaI toxicity between 24hr and 48hr. The LD_(50) were 84. 05μgand 58.64μg total protein/ml in vitro after 24hr and 48hr respectively.The larvicidaltoxin from B. t.may facilitate microbial control of parasitic nematodes.
