高温作业工人血清抗HSP_（70）抗体水平的研究

摘　　要：本研究用免疫印迹技术，以纯化的ＨＳＰ７０为抗原，检测了高温作业工人血清抗ＨＳＰ７０抗体的水平。实验发现；用人体来源的ＨＳＰ７０作为抗原的检测比动物（大鼠）来源的ＨＳＰ７０作为抗原的反应灵敏得多。用人体细胞制备的ＨＳＰ７０作为抗原时，１０例高温作业工人中有２例呈阳性反应，而非高温对照１０人中仅１例呈弱阳性反应，含此抗体阳性血请用动物来源的ＨＳＰ７０作抗原时显著减弱。用动物来源的ＨＳＰ７０作抗原，检测５０例不同时象（以高温工人对比，白血病、心肌炎、肝癌等患者和正常对照者）的血清，除２例高温工人呈阳性反应外，余均为阴性。动物与人体来源的ＨＳＰ７０免疫源性差异，可能是产生不同结果的主要原因。通过本研究，我们认为用纯化的ＨＳＰ７０或其他ＨＳＰｓ作为抗原来检测人体抗ＨＳＰｓ抗体的方法是可行。n this experiment,anti-HSP70 of serum in different groups such as workers working inthe extremely hot enviroment,students , patients with leukermia , liver cancer and heart dis-ease were detected with Western blot using purified HSP70. The results shows that there ex-isted a difference when using HSP70 antigen of different origins using human HSP70 as anti-gen,2 obvious positive stainings were observed in 10 workers working in the very hot enviro-ment and I in 10 students. Using animal-original HSP70 in 50 human sera,only 2 workersshowed as positive in workers working in the very hot enviroment. Unfortunately,we havenot yet investigated the characters of these two individuals. Anyway, this result offered apo.sible way for large-scale immunological investigation.
