摘  要:本文对甘薯黑斑病菌抗性差异显著的两个甘薯品种鄂薯1号(高抗)和徐薯18(高感)在接种黑斑病菌孢子悬浮液初期进行了其脂质过氧化代谢规律的研究。从脂质过氧化物含量和脂氧合酶活性分析表明,在接种初期,抗病品种鄂薯1号比感病品种徐薯18发生了较强的脂质过氧化作用,揭示了脂质过氧化作用介入了甘薯对黑斑病菌的抗性机制。进一步从鄂薯1号块根组织中利用Sephadex G—200柱凝胶层析分离纯化了脂氧合酶,酶活性提高8.9倍,回收率达25%,通过Lineweaver—Burk作图测定其Km值为1.2mmol/l,经SDS—聚丙烯酰胺凝胶电泳测定其分子量为89.1KDa。The Lipid peroxidation in sweet potatoes was studied. The content of lipid peroxides and the activity of Lipoxygenase in the high resistant cultivar Eshu- 1 were much higher than that of the sensitive cultivar Xushu- 1 8 at the early period after they had been infected by Ceratocystis fimbriata. A much more details of lipoxygenase in Eshu- 1 was studied, the activity of lipoxygenase increased 8. 9- fold and was obtained recovery of 25 percent after purified by Sephadex G-200 column, the Km value of lipoxygenase is 1. 2 mmol/1 determined by plot of Lineweaver- Burk and its molecular weight was 89. 1 KD by SDS-polyacrylamide gel electrophoresis.
