摘  要:用亲和层析法从3-5月龄引产胎儿肌肉组织中提纯AFP。体外用~3H-UR参入的方法,从转录水平观察AFP对小鼠H_(22)腹水型肝癌细胞生长的影响。结果表明,人AFP能显著地直接促进该肿瘤细胞RNA的合成;人血白蛋白和去除AFP后胎儿肌肉匀浆液等对照均无显著作用;AFP抗血清能显著地清除AFP的这种促进作用。用小鼠S_(180)乳腺肉瘤细胞作为对照表明,AFP促肿瘤生长无严格的组织特异性。human Alpha Fetoprotein (AFP) was purified from fetus by means of affinity chromatography and its effect on mouse hepatoma-22(H-22) ascites cell was observed by the ~3H-UR incorporation technic. The results showed that AFP stimulated the RNA synthesis of H-22 ascites cells significantly.This effect seemingly resulted from direct action of AFP on H-22 ascites cells rather than indirect suppression of immune system by AFP. Human serum albumin and the feto-homogenates free from AFP had no effect on the RNA synthesis of H-22 ascites cells. On the other hand the effect of AFP was significantly abolished by the antiserum of AFP. Compared with the reactive system of mouse Sarcoma-180 cells, this effect of AFP showed no strict tumor specificity.
