色素痣和黑素瘤的DNA含量与病理形态学及预后的关系

摘  要:应用流式细胞术(FCM)定量研究25例PN、29例MM及8例皮肤的DNA含量和细胞增殖特性。结果表明MM的SPF和PI显著高于PN和正常皮肤(P<0.01),二倍体MM的SPF和PI明显高于PN(P<0.05),DI、PI与MM组织类型部分有关,MM的核分裂指数和血管数目与DI、PI呈正相关(P<0.05)。提示黑素瘤的FCM检测可以鉴别其良恶性,在一定程度上与其组织病理指标相关。Nuclear DNA content and cellular proliferationfeatures of 25 cases of pigmented nevus (PN), 29 malignant melanomas (MM) and 8 normal skin for controlwere quantitatively studied by flow cytometry (FCM).correlations between nuclear DNA content and histological type biological behavior in PN and MM were analyical. All normal skin tissues were DNA-diploid, 21 of25 PN were DNA-diploid, 4 were near diploid andaneluploid (16%). 25 of 29 MM were aneuploid(86%). The aneuploid rate in MM was significantlyhigher than that of PN (P<0. 01 ). S-phase fraction(SPF) (19. 25±3. 41 ) and proliferation index (PI) (33.55±6. 81) in MM were markedly higher that these ofcontrol (8. 33±1. 97; 10. 78±3. 38),and PN (12. 78±3. 72; 21. 63±6. 76), respectively (P < 0. 01; P< 0.01),SPF and PI in MM were notably higher in diploidthan in PN of diploid and near diploid (P<0. 05), Itsuggested that aneuploid may be regarded as a markerof MM:SPF and PI may be veluable in reflecting themalignant degree and growth behavior of tumors and indistinguishing benign from malignant tumor. From thisstudy we suggest that FCM-DNA may be a valuable indicator in differential diagnosis and prognosis of benignand malignant melanocytic tumors.
