应用MTT比色法测定EPO体外生物学活性

摘　　要：EPO依赖细胞珠在EPO浓度梯度中培养72小时，再加入MTT培养5小时，离心弃上清后，加入有机溶剂溶解MTT代谢产物，以OD值对EPO浓度梯度作量效曲线，定量地测定EPO生物学活性。本法重复性好，与其它体外生物学活性测定方法所测定结果相符。EPO dependent cell strains cultured in EPO concentration gradient for 72hours were continuously culttired for 5 hours after MTT were added. The MTT metabolicproducts were separated by centrifugation and dissolved by organic solvents, then detectedbyspectrophotometry. Dose-effect graph was made by plotting EPO concetration against ODvalue. and used for the quantitative determination of the biological activity of EPO. Themethod showed good reproducibility compared with other methods.
