多粘菌素B对S_（180）细胞增殖和膜脂肪酸组成的影响

摘　　要：观察到环肽类抗生素多粘菌素Ｂ对小鼠肉瘤Ｓ１８０细胞增殖具有显著抑制作用，半数有效抑制浓度（ＩＣ５０）为８．９５×１０５ＩＵ·Ｌ－１。用毛细管柱气相色谱法分析了多粘菌素Ｂ作用于Ｓ１８０细胞２４ｈ后膜脂肪酸组成的变化，发现磷脂的花生四烯酸（Ｃ２０：４）显著减少，中性脂的硬脂酸（Ｃ１８：０）增多，油酸（Ｃ１８：１）减少，硬脂酸与油酸的比值（Ｃ１８：０／Ｃ１８：１）升高．提示多粘菌素Ｂ对Ｓ１８０细胞增殖的抑制与膜脂肪酸组成的变化有关。Abstract The effect of polymyxin B (PMB)on the proliferation of S180 cells was studied. PMB was able to inhibit the cell proliferation obviously and the IC50was 8.95 ×105 IU·L-1.Toinvestigatethemechanism of antitumor action of PMB. the fatty acid composition of phospholipids and neutral lipids of S180 cell membranes was analysed by capillaries gas chromatography after PMB acted on cells for 24 hours. The results showed that the arachidonic acid ( C20:4) of phospholipids reduced significantly ; the oleic acid (C18: 1) reduced; but the stearic acid ( C10:0) and the ratio of C18:0/C18:1increased(P<0.01) .The data suggest that the inhibitory effect of PMB on the proliferation of S180 cells may be related to the changes of fatty acid composition.
