摘 要:为探讨冲胚方法对小鼠超数排卵效果的影响,分别采用子宫切碎法与子宫冲胚法对超排小鼠进行了冲胚.结果表明:子宫冲胚法的平均采卵数和平均获胚数极显著高于子宫切碎法(P<0.01),而平均可用胚数显著高于子宫切碎法(P0.05),说明冲胚方法对小鼠超数排卵效果的影响显著,且子宫冲胚法明显优于子宫切碎法.小鼠早期胚胎发育观察结果表明:超排处理后,在公母鼠合笼后76~79.5 h采胚,胚胎大多处于桑椹胚期,而在公母鼠合笼后88~91.5 h采胚,胚胎大多处于致密桑椹胚期与囊胚期;采用TCM199+15%胎牛血清培养液对小鼠胚胎进行体外培养是可行的.In this paper,the effect of different methods of collecting embryos on the superovulation of mice was studied,and the embryos in the superovulated mice were collected by using the uterus-mincing method and uterus-collecting method respectively.The results showed that the average number of eggs and embryos obtained by the uterus-collecting method were extremely significantly higher(P<0.01) than those obtained by the uterus-mincing method,and the average number of usable embryos obtained by the uterus-collecting method was significantly higher(P<0.05) than that did by the uterus-mincing method.The average number of unfertilized eggs obtained by these two methods was not significantly different(P>0.05).So the superovulation effect of mice was significantly influenced by the method of collecting embryos.The observation results of early embryos development indicated that most of the embryos collected 76 ~79.5 hours after combining cages were at morula stage,while most of the embryos collected 88 ~ 91.5 hours after combining cages were at compacted morula stage and blastocyst stage.It was feasible that the embryos of mice were cultivated in vitro by using the mixed solution of TCM199 and 15% FCS.
