摘  要:目的:检测白细胞介素(IL)-29对肥大细胞Th2细胞因子分泌的影响和可能的信号转导通路。方法:肥大细胞P815培养、激发后收集不同时间点的细胞和上清液,用酶联免疫吸附试验(ELISA)检测上清液中IL-4和IL-13的水平,用细胞激发信号ELISA(CASE)方法检测丝/苏氨酸蛋白激酶(Akt)、细胞外信号调节激酶(ERK)、信号转导子和转录激活子(STAT)3和p38丝裂原活化蛋白激酶信号转导通路蛋白磷酸化情况。结果:IL-29能够促进肥大细胞P815分泌IL-4和IL-13。AG490和LY294002阻断IL-29引起的肥大细胞IL-4分泌并抑制IL-29引起的STAT3和Akt磷酸化。LY294002阻断IL-29引起的肥大细胞IL-13分泌并抑制IL-29引起的Akt磷酸化。结论:IL-29刺激肥大细胞P815分泌IL-4可能是通过激活Akt和STAT3信号转导通路实现的,而IL-29刺激肥大细胞P815分泌IL-13可能与Akt信号转导通路激活有关。Objective:To investigate the potential influence of interleukin(IL)-29 on mast cell IL-4 and IL-13 release as well as the potential signal transduction pathways correspondingly.Methods:After P815 mast cells were challenged with different concentrations of IL-29,the supernatant of challenged cells was analyzed by enzyme-linked immunosorbent assay(ELISA) to detect the release of Th2 cytokines,IL-4 and IL-13,and the challenged cells were treated and analyzed by cellular activation of signal ELISA(CASE) to detect phosphorylation of serine/threonine protein kinase(Akt),extracellular signal-regulated kinase(ERK),p38 mitogen-activated protein kinase(MAPK) and signal transducer and activators of transcription 3(STAT3).Results:Both AG490 and LY294002 can not only abolish IL-4 release from mast cells induced by IL-29,but also inhibit phosphorylation of STAT3 and Akt induced by IL-29.At the same time LY294002 can block IL-13 release induced by IL-29 as well as phosphorylation of Akt.Conclusion:Activation of Akt and STAT3 signalling pathways may be involved in IL-4 release from mast cells induced by IL-29,whereas IL-29 initiated IL-13 release may be related to activation of Akt signalling pathways in P815 mast cells.
