芒果苷对化疗荷瘤小鼠细胞因子及T细胞内第二信使水平的影响

摘　　要：目的:探讨芒果苷对化疗荷瘤小鼠外周血细胞因子白介素-2(IL-2)和肿瘤坏死因子-α(TNF-α)及脾T淋巴细胞内第二信使环腺苷酸(cAMP)的影响。方法:建立肉瘤S180荷瘤小鼠模型,并使用环磷酰胺(Cyclophosphamide,CTX)及CTX联合芒果苷对其进行治疗,采用HE染色法检测肿瘤组织的病理变化;ELISA法测定治疗后小鼠外周血细胞因子IL-2、TNF-α;酶联免疫斑点法(ELISPOT)检测脾T淋巴细胞内第二信使cAMP的水平。结果:芒果苷具有联合CTX抗肿瘤作用,可拮抗CTX对荷瘤小鼠的毒副作用;能升高血清中IL-2的水平,提高TNF-α的水平;降低脾T淋巴细胞内cAMP的水平。结论:芒果苷可拮抗CTX对机体的毒副作用,维持机体的免疫,联合使用可增强化疗药物的抗肿瘤效果。Objective:To investigate the effects of mangiferin on the levels of interleukin(IL)-2,Tumor necrosis factor(TNF)-α from peripheral blood,and cAMP from spleen T lymphocytes in carcinomatous mice after chemotherapy.Methods:The mice models with tumor S180 were established and treated respectively by cyclophosphamide(CTX) and CTX combined with mangiferin.The levels of IL-2 and TNF-α in mice peripheral blood were determined with ELISA;the contents of cAMP in spleen T lymphocytes were detected by ELISPOT.Results:Mangiferin can antagonize the toxicity of CTX on tumor-bearing mice,maintain the weight,spleen index and thymus index of tumor-bearing mice;promote the content of IL-2;stabilize the level of TNF-α and reduce the content of cAMP in spleen T lymphocytes.Conclusion:Mangiferin can antagonize the toxicity of CTX on the body,maintain the normal immunity and enhance the effect of CTX on anti-tumor.
