自体成纤维细胞注射移植对衰老模型鼠皮肤结构的影响

摘　　要：目的:探讨自体成纤维细胞注射移植对衰老模型鼠皮肤结构的影响。方法:切取大鼠背部全层皮肤培养成纤维细胞,同时给大鼠注射定量的D-半乳糖促使其衰老即建立衰老模型,最后在衰老模型上注射移植培养的自体成纤维细胞,用显微镜观察鼠皮肤组织切片,图像分析仪计算胶原面积比值和测量真皮及皮下组织的厚度,并对所得数据进行统计学分析。结果:移植的成纤维细胞存活;细胞注射1个月时,实验组和对照组的胶原面积比值和真皮厚度相比差异无统计学意义(P>0.05),2个月时,实验组和对照组的胶原面积比值和真皮厚度相比差异有统计学意义(P<0.01)。结论:移植的自体成纤维细胞能够存活并分泌胶原细胞,增加皮肤厚度,使皮肤外观丰满。Objective:To investigate the effect of autologous fibroblast transplantation on skin structure in rat aging model.Methods:Cutting back full-thickness skin of rat to cultivate fibroblasts was performed,at the same time injecting quantitive D-galactose into rat to establish aging model was done,eventually transplantating autologous fibroblasts into aging model was completed.Rat skin was brought out to the heredity substance and observed by microscopy.The collagenic area and dermal thickness were measured with image analysis software,and statistical analysis was also carried out.Results:Transplanted fibroblasts could survive.Both the area ratio of collagen fibers and the dermal thickness were significantly increased in injected cells after 2 months(P0.05).Conclusion:Autologous cultured fibroblast could survive after transplantating into the skin and collagen could be newly produced,the depth of dermis increases which provides a possibility to treat mini-defects of skin.
