摘 要:目的:观察桔梗对异烟肼(INH)和利福平(RFP)合用致小鼠肝损伤的保护作用,并探讨其作用机制。方法:小鼠灌胃给予异烟肼和利福平(各100 mg.kg-1)制备肝损伤动物模型,同时分别灌胃给予低、中、高剂量桔梗(1.5、2.0、2.5 g.kg-1),14 d后,测定小鼠血清中ALT含量、肝脏指数以及肝组织匀浆中的MDA、SOD的活性,观察肝组织病理学切片。结果:桔梗能降低肝脏指数(P<0.05)、降低小鼠血清ALT含量、降低小鼠肝匀浆中的MDA、增加肝匀浆中SOD值(P<0.05),减轻肝组织变性、坏死程度,缓解肝组织的病理改变。结论:桔梗对INH和RFP合用所致的小鼠肝损伤具有保护作用,其作用机制可能与桔梗的抗脂质过氧化作用有关。Objective: To observe the protective effect of platycodon grandiflorum on mice hepatic toxic injury induced by isonicotinic acid hydrazide(INH)and rifampin(RFP)when used in combination.Methods:The hepatic injury model in mice was produced by intragastric administration with INH(100 mg·kg-1) and RFP(100 mg·kg-1).And Platycodon grandiflorum was given by intragastric gavage with the dose of(1.5,2.0,2.5 g·kg-1).The activity of ALT in serum,liver index,contents of MDA and activity of SOD in liver,histopathologic changes of liver and ultramicrostructure of hepatocyte were detected in 14 days.Results:Compared with the model group,platycodon grandiflorum could reduce the liver index,inhibite raising of activity of ALT in serum(P<0.05),and make the content of MDA reduced and the content of SOD induced in the hepatic tissue.In the experimental group were hepatic cellular swelling,fatty degeneration,spotty necrosis,and the lipid droplet in hepatocyte relieved.Conclusion:The protective effect of platycodon grandiflorum on mice hepatic injury induced by both INH and RFP may be related to stabilizing the liver membrane and inhibiting the lipid peroxidation.
