摘  要:目的:实低温处理成纤维细胞复苏过程存在延迟再水化现象。方法:-10℃处理成纤维细胞后20℃复温,培养1d后离心收集上清漂浮细胞培养,观察脱水漂浮细胞再次贴壁、检测细胞细胞贴壁率、分裂指数、用噻唑蓝(MTT)法确定细胞存活来确证漂浮细胞可以延迟再水化。结果:部分漂浮细胞更换培养瓶后,延迟水化,再次贴壁,检测代谢指标显示细胞代谢功能恢复。讨论:冻伤后部分细胞由于脱水出现休眠状态,予以合适条件后细胞可以再次复苏、水化。Objective:To verify the phenomenon of delayed rehydration of normal human fibroblasts after freezing and thawing. Methods: Samples were cooled in a rate of 1 0. 1℃/ min down to the experimental temperature of - 10℃, and they were warmed at 20℃. After being cultured for one day the cells suspending in the medium were collected and seeded. The adherent rate and mitotic index of the fibroblast were assessed. MTT test -was used to verify the phenomenon of rehydration of survived fibroblasts. Result: After freezing and thawing, the fibroblasts still had ability to adhere. MTT test showed that the cells survived. The experiment verified the phenomenon of delayed rehydration of normal human fibroblasts after freezing and thawing. Conclusion: Rehydration is a kind of protective mechanism,and when the offence of low temperature is removed, the cell might revive.
