摘 要:目的探讨七叶皂甙钠对大鼠大脑中动脉闭塞缺血再灌注后缺血区神经元凋亡的影响。方法选用健康雄性Wistar大鼠140只,用线栓法建立大鼠大脑中动脉缺血再灌注模型,切取不同时间段缺血半暗带和中心区皮质组织,采用逆转录聚合酶链反应(RTPCR)测定凋亡相关基因(Fas、FasL与Bcl2)转录水平的变化;干预组再灌注即刻开始腹腔内注射七叶皂甙钠(5mg/kg),以后5mg·kg1·24h1治疗量,而对照组再灌注即刻注射等量生理盐水;对选取的脑组织块行原位末端转移酶标记(TUNEL),计数缺血半暗带和中心区凋亡细胞数目。结果干预组各时段大鼠脑缺血组织神经元凋亡计数明显低于对照组(P<0.01),表达高峰下降明显,干预组脑缺血组织中Fas、FasLmRNA表达明显下调,同时可见Bcl2mRNA表达上调。结论七叶皂甙钠具有抗神经元凋亡作用;调节神经元凋亡相关基因表达水平可能是其机制之一。Objective To observe the effects of sodium aescinate on neuronal apoptosis' gene in acute middle cerebral artery occlusion and ischemia-reperfusion model of rats. Methods The middle cerebral artery occlusion models were established by using the intraluminal suture occlusion method in 140 Wister rats. The rats were killed to observe the neuronal apotosis' gene (Fas,Fas-L and Bcl-2) levels of ischemia penumbra and cora central tissue at different time by using reverse transcription-polymerase chain reaction (RT-PCR). Injected rats in sodium aescinate group were reperfused and sodium aescinate (5 mg/kg) was abdominally injected at the time, and then sodium aescinate (5 mg/kg) was abdominally injected every 24 hours. The rats in contral group were injected the same doses saline. The neuronal apoptosis cells in rat ischemia penumbra and cora central tissue were evaluated by TUNEL assay. Results In injected sodium aescinate group, apoptosis positive cells mainly distributed in ischemia zone at different time (P〈0. 01) and the peak of expression diminished obviously. In injected sodium aescinate group, Fas,Fas-LmRNA was in few expression in ischemia tissue, and the overexpression of Bcl-2mRNA was detected in ischemia zone. Conclusions Sodium aescinate has an effect on neuronal apoptosis and the mechanism of nural protection may be involved with regulation of neuronal apoptosis' gene expression.
