摘  要:目的研究葛根素对大鼠全脑缺血再灌注损伤的保护作用及其机制。方法采用4血管闭塞法建立SD大鼠全脑缺血再灌注模型,并给予葛根素干预,于缺血再灌注后6、24、72h、5、7d进行HE染色及免疫组织化学染色,分别检测大鼠脑组织海马CA1区神经元数目、形态变化和bc12表达。结果(1)脑缺血再灌注后6、24h海马CA1区神经元未见明显死亡,再灌注后72h、5、7d葛根素组神经元存活数目显著高于无药物干预的再灌注组;(2)葛根素组与再灌注组比较各时间点bc12表达均明显增多;(3)葛根素组与再灌注组神经元存活数目与bc12阳性细胞数均呈显著正相关。结论葛根素对脑缺血再灌注损伤有保护作用,可延缓迟发性神经元死亡的发生,其机制可能与葛根素上调抗调亡基因bc12表达有关。Objective To investigate the protective effects and mechanisms of puerarin after global cerebral ischemic reperfusion in rat. Methods 4-vessel occlusion method was used to establish the models of global cerebral ischemic reperfusion in Sprague-Dawley(SD) rats. At the respective time points at 6h, 24h,72h, 5d and 7d after ischemia and reperfusion, hematoxylin-eosin staining was applied to observe neuronal pathomorphological changes and the number of living neurons in hippocampus CAI region, and immunhistochemical method was performed to determin the positive expression of bcl-2. Results (1) Few neurons in hippocampus died at 6h and 24h after ischemia and reperfusion. At 72h,5d and 7d the number of living neurons in puerarin group was significantly more than that of the nontreated ischenmia-reperfusion group; (2) The number of bcl- 2 positive cells in the puerarin group significantly increased compared to that in ischemia-reperfuson group at respective time; (3) The positive correlation between the number of living neurons and the number of bcl-2 positive cells in hippocampus CAI was significant in both groups. Conclusions Puerarin can protect rat cerebral ischemia-reperfusion injury and reduce delayed neuronal death, and this may relate to its effect of upregulating the expression of bcl-2.
