摘  要:目的探导可溶性鸟苷酸环化酶(sGC)α1、β1亚基蛋白在大鼠脑缺血再灌注后血管平滑肌中的表达及其意义。方法将SD大鼠72只随机分为颈内动脉和大脑中动脉两组,并又随机分别分为对照和缺血两亚组。缺血亚组分别手术夹闭颈内动脉或大脑中动脉0.5、1、2、2.5h,再灌流至24h,处死动物。取夹闭部远端动脉行免疫组化染色,用吸光度分析软件读取动脉血管的平均吸光度值。分别计算各组大鼠脑血管中sGCα1、β1亚基蛋白的含量。结果对照组大鼠颈内动脉和大脑中动脉中膜平滑肌sGCα1、β1亚基蛋白呈强阳性反应,平均吸光度值分别为0.17±0.13和0.18±0.17;颈内动脉夹闭0.5、1、2、2.5h组平均吸光度值分别为0.15±0.11、0.13±0.13、0.14±0.09和0.15±0.12,大脑中动脉夹闭0.5、1、2、2.5h组分别为0.15±0.16、0.12±0.09、0.08±0.03和0.06±0.02。在夹闭大脑中动脉后2、2.5h平均吸光度值明显高于对照组(P<0.01)。结论sGCα1、β1亚基蛋白表达减少及功能的下降,可减小内源性和外源性NO的反应能力,从而抑制损伤血管新生内膜的形成,对脑组织损伤具有保护作用。Objective To explore the expressions of α1 and β1 subunits of the soluble guanylate cyclase (sGCα1,β1) in the vascular smooth muscle after cerebral ischemia-reinfusion in rats and their significance. Methods Seventy-two SD rats were randomly divided into two groups, i.e. the group of occluding the internal carotid artery (ICA) and the group of occluding the middle cerebral artery (MCA). The each group was divided again randomly into control subgroup (n=4) and ischemia-reinfusion subgroups (24 hours reinfusion at 0.5, 1, 2 and 2.5 hours respectively after ischemia, n=8). The expressions of sGCα1 and β1 in the vascular smooth muscle distal to the occlusion point were determined by immunohistochemical technique. The mean absorbance (MA) was read by MA analysis software. The contents of the sGCα1 and β1 in the cerebral arteries were calculated in each group. Results In the control group, the expressions the sGCα1 and β1 subunits in the smooth muscle of the ICA and MCA were strongly positive (MA was 0.17±0.13 and 0.18±0.17 respectively). The MA 0.5, 1, 2, 2.5 hours after the ICA occlusion was 0.15±0.11, 0.13±0.13, 0.14±0.09 and 0.15±0.12 respectively, and 0.5, 1, 2, 2.5 hours after in the MCA occlusion was 0.15±0.16, 0.12±0.09, 0.08±0.03 and 0.06±0.02 respectively. The MA in 2 and 2.5 hours occlusion of MCA subgroup was significantly lower than that in the control group (P〈0.01). Conclusions The decrease in sGCα1 and β1 expressions and their function can lower the reaction to the internal and external NO, thus suppressing the formation of neointima in the injured vessle, and therefore has protection against the cerebral injury.
