摘 要:目的改进大鼠大脑中动脉梗塞法,建立更接近于临床缺血性脑卒中及其再灌注的可靠模型.方法沿大鼠右颈外动脉入栓,经颈内动脉到达大脑中动脉,入栓线长15~18mm,直径0.205mm,阻断其血流,观察大鼠神经病学改变及脑组织形态学变化.结果术后大鼠表现出特殊体态及典型追尾征,6h后大脑中动脉供血区出现缺血性外观(TTC染色)及相应组织学变化(HE染色),模型成功率近83%.结论本方法既可构成缺血性病灶,又能形成再灌注模型,具有可逆性重复操作的特点,其神经病学与形态学的特殊表现,可作为鉴定模型的可靠依据.Objective To establish a more ideal animal model of middle cerebral artery occlusion (MCAO). Methods In rats,a fine and glossy nylon thread which was 0. 205mm in diameter was introduced into the right cervical internal carotid artery, till the intromitting length equal 15 - 18mm to block blood flow of MCA, then the changes of neurological syndromes and morphology of brain tissue were observed. Results After MCAO, special neurological syndromes were showed by rats, the infarct surface in the area of MCA ( TTC coloring, after MCAO 6h) and corresponding changes of brain tissue ( HE staining) were appeared. The success rate of the model reached 83%. Conclusion A reversible model of regional cerebral ischemia in rats was developed, the reliability of which could be evaluated by neurological syndromes.
