摘  要:目的研究针刺不同经腧穴对偏头痛大鼠脑干组织G蛋白亚基(Gsα和Giα)含量的影响。方法通过皮下注射硝酸甘油(10mg/kg)建立实验性偏头痛大鼠模型,将动物随机分成正常对照组、生理盐水组、模型对照组、针刺肝经组、针刺胆经组和针刺三焦经组,运用免疫印迹法(Westernblot)检测脑干组织Gsα和Giα的含量。结果皮下注射硝酸甘油4h后脑干组织Gsα蛋白含量明显升高(P<0.01),Giα蛋白含量明显降低(P<0.01),Gsα/Giα蛋白比值升高;三个针刺治疗组与模型组比较,脑干组织中Gsα蛋白含量明显降低(均P<0.01),Giα蛋白含量明显升高(均P<0.01),Gsα/Giα蛋白比值降低;而针刺胆经组G蛋白含量变化最为明显。结论偏头痛发作可能与大鼠脑干组织G蛋白信号传导系统功能障碍有关;针刺介导的G蛋白信号通路可能是其防治偏头痛的重要机制之一;针刺胆经腧穴较针刺其他两组经腧穴介导G蛋白信号通路作用更强。[Objective] In order to explore the effect of acupuncture the different meridian points on the content of G protein alpha subunits in the migraine rat brain stem after nitroglycerin infusion. [Methods] The model of migraine rats were reproduced in accordance with Knynihar-tassorelli methods by infusing the NO donor nitroglycerin [glyeeryl trintrate (GTN)] i.h, then the rats were divided randomly into six groups: normal control group, saline control group, model control group and acupuncture liver meridian group, acupuncture gallbladder meridian group, acupuncture san jiao meridian group. The content of Gsα and Giα protein in the rat brain stem was analysed by western blot method. [Results] The model control group of the content of Gsα protein in the rat brain stem was signifieantly raised (P 〈0.01), the Giα protein was significantly reduced (P 〈0.01), and the ration of Gsα/Giα was increased by comparison with the normal control group. The three acupuncture groups of Gsα protein in the rat brain stem went down apparently than the model group (P 〈0.01), and Giα protein was slightly higher than the model group (P 〈0.01), resulting in reducing of Gsα/Giα rations, and the acupuncture gallbladder channel group have the most change in the three treatment groups. [Conclusion] The results provided important information for understanding how G-protein-linked signaling system play an important role in the attacking migraine, and the mechanisms of acupuncture on migraine was possibility that the acupuncture mediated the G-protein-linked signal transduetion pathway plays an important function. Stimulating the gallbladder meridian points can mediate the G-protein-linked signal transduetion pathway more significantly than the other two acupuncture treatment groups.
