实验性脑出血后细胞凋亡的变化及依达拉奉的干预作用

摘　　要：目的:研究依达拉奉对脑出血后神经功能缺损、细胞凋亡及caspase-3表达的影响。方法:SD大鼠分为正常组、假手术组、模型组与治疗组,后2组分为6h和1、3、5天4个亚组,每组5只,模型组与治疗组采用Ⅶ型胶原酶诱导脑出血模型,假手术组以等量生理盐水代替Ⅶ型胶原酶,治疗组予以依达拉奉3mg/kg腹腔注射,每天1次。于6h和1、3、5天观察大鼠神经行为学评分,应用TdT介导的原位末端标记(TUNEL)法标记断裂的DNA片段检测凋亡细胞,免疫组化方法测定caspase-3阳性细胞数。结果:与模型组比较,依达拉奉干预后3天和5天大鼠神经行为学评分明显减少(P<0.05),1、3、5天时caspase-3阳性细胞数明显减少(1天和3天P<0.05,5天P<0.01),TUNEL阳性细胞数于3天和5天时明显减少(3天P<0.05,5天P<0.01)。结论:依达拉奉能有效减轻脑出血后的细胞凋亡,改善神经功能缺损症状。Objective： To study the effect of edaravone on neurological function deficit,neuron apoptosis and expression of caspase-3 in rats after ICH. Methods： All rats were randomly divided into normal group,sham-operation group,model group and treatment group.The later two groups were divided into four subgrnups respectively ： 6-hour group, l-day group,3-day-group and 5-day group with （each group 5 rats ）. We established ICH model in model group and treatment group by injection of collagenase Ⅶ. Shamoperation group was established by saline instead. Treatment group were injected edaravone 3 mg/kg through celiac once a day.At 6 hour, 1 day,3 day and 5 clay.the score of neurological behavior in rats were observed,TUNEL was used to detect deoxyribonucleic acid （DNA） fragmentation in apoptosis cell,caspase-3 positive cell were measured by immunohistochemistry. Results： Contrast to model group,the score of neurological behavior in treatment group rats were decreased significantly at 3 day and 5 day （P 〈 0.05）,as well as TUNEL positive cell （3 d P 〈 0.05,5 d P 〈 0.01 ）.Significant difference of caspase-3 positive cell between model group and treatment group occurred at 1 day,3 day and 5 clay （ 1 d,3 d P 〈 0.05,5 d P 〈 0.01 ）. Conclusion： Edaravone can effectively alleviate apoptosis and accelerate the recover of neurological function deficit after ICH.
