摘  要:目的探讨人类转移抑制基因KiSS1及其受体hOT7T175在乳腺癌中的表达情况及临床意义。方法以逆转录聚合酶链反应(RTPCR)方法检测40例癌组织和癌旁正常组织中KiSS1mRNA和hOT7T175mRNA的表达。结果40例乳腺癌组织中,17例(42.5%)KiSS1基因表达阳性,10例(25%)hOT7T175基因表达阳性;正常组织中29例(72.5%)KiSS1基因表达阳性,20例(50%)hOT7T175基因表达阳性。KiSS1基因和hOT7T175基因失表达与临床分期、肿瘤大小等均无显著相关性,而KiSS1基因在淋巴结阴性者表达阳性率为63.6%,阳性者仅为16.7%,两者差异有统计学意义(P<0.01)。结论KiSS1基因失表达与乳腺癌的淋巴结转移密切相关,提示该基因的表达可能是乳腺癌中一个重要的监控淋巴结转移的生物学标志。Objective To investigate the exprssion of KiSS-1 and hOT7T175 gene in breast cancer and its clinical significance. Methods The mRNA expression of KiSS-1 and hOT7T175 gene was detected in benign and malignant breast tissues by RT-PCR in 40 patients. Results In 40 cases of breast cancer tissues, the positive rate of KiSS-1 gene and hOT7T175 gene expression was 42.5% and 25 % respectivel); while in normal tissue the rate was 72.5% and 50% respectively. Loss of KiSS-1 and hOT7T175 gene expression in breast cancer was not correlated with clinical stage, tumor size. The positive KiSS-1 rate was 63.6% in those without lymph node metastasis, but only 16.7% in those with metastasis ( P〈0.01 ). Conclusion Loss of KiSS-1 gene expression may be a useful marker for detection of lymph node metastasis in breast cancer.
