摘  要:目的观察外源性非酶糖基化终末产物(AGEs)在视网膜中对血管内皮生长因子(VEGF)受体(Flt-1)的表达和对视网膜血管的影响。方法应用鼠血清白蛋白(RSA)体外孵育AGEs,并于尾静脉注射至健康大鼠体内,每日1次,连续2周,随机分为AGEs组和RSA对照组。通过眼底荧光血管造影(FFA)观察视网膜血管有、无渗漏,采用Envision系统免疫组织化学法检测视网膜Flt-1的表达,通过视网膜超薄切片和透射电镜观察视网膜血管超微结构改变。结果2周后FFA显示大鼠视网膜均未出现荧光渗漏、积聚现象,未发现有毛细血管无灌注区。视网膜内核层中出现棕黄色Flt-1阳性染色细胞,AGEs组的阳性细胞[(25.18±3.44)个]较RSA对照组多50%[(18.74±4.11)个,P<0.05]。AGEs组大鼠视网膜毛细血管基底膜电子密度轻度增高,周细胞、内皮细胞胞浆内线粒体发生肿胀、膜破裂;RSA对照组未发生超微结构的改变。结论AGEs可作为独立因素参与视网膜血管及其细胞的损害过程,并与VEGF有一定的联系。Objective To observe whether advanced glycation end products(AGEs) have influence on vascular endothelial growth factor (VEGF) receptor flt-1 expression and retinal vasculature. Methods Healthy rats were infused with AGEs derived from RSA or native RSA as control. Fluorescence fundus angiography(FFA), immunochemistry with anti-fit polyclonal antibody, retina ultrathin sections and transmission electron microscopy were performed. Results Retinal vessels showed absence of dye leakage or accumulation. Positive brown cells for flt-1 were prominent in the inner nuclear layer. AGEs group had 50% positive cells more than the control group(AGE-RSA vs RSA, 25.18±3.44 vs 18.74±4.11, P〈0.05). The retinal capillary vessel basement membrane showed slightly increased electron density with mitochondrial swelling of pericyte and endothelium in the AGE infusion group, whereas the RSA control group did not. Conclusion The AGE plays an independent role in the impairement course of retinal vessels and cells, having some relations with VEGF. (Shanghai Med J, 2005, 28:672-674)
