苯并(a)芘对小鼠肺组织细胞色素P450 1A表达的影响

摘　　要：目的研究苯并(a)芘(BaP)作用下小鼠肺组织细胞色素P4501A(CYP1A)的表达和肺组织病理形态学改变。方法昆明种雄性小鼠56只,随机分为2组(28只/组),BaP染毒组(4个亚组,7只/亚组),分别以0、0.32、1.58、7.89mg/kg体重腹腔注射BaP,每周染毒4d,持续7周;预热染毒组(4个亚组,7只/亚组),每次实验前39℃,预热2h,再用染毒组相同的剂量和时间至染毒结束。其中0mg/kg体重BaP为植物油溶剂;采用West ernblot法检测小鼠肺组织CYP1A表达水平,光镜观察肺组织病理学变化。结果0.32mg/kg体重BaP染毒时,小鼠肺组织CYP1A表达增加,BaP剂量继续增加,CYP1A表达反而降低;预热应激组CYP1A表达有相同的变化趋势,但比相同剂量的BaP染毒组表达水平高;BaP染毒后小鼠肺组织有炎性增生的表现。结论低剂量BaP染毒诱导小鼠肺组织CYP1A表达增加,高剂量BaP染毒则抑制CYP1A表达。Objective To study the changes of the cytochrome 1A （CYP1A） expression and pathologic morphology in lung tissue of mice treated with benzo （a） pyrene （BaP） . Methods Fifty-six male Kunming mice were averagely divided to two groups. In the BaPgroup, the mice were subjected to the intraperitoneal injection of different concentrations of BaP （0, 0.32, 1.58, 7.89mg/kg weight） 4 times each week for 7 weeks continuously. In the preheated group, the mice were heated by 39℃ for 2h first, then treated by the same BaP concentrations as in BaP groups. The CYP1A protein expression of lung tissue was detected by using Western blot, and the pathologic changes were observed under the microscopy. Results The CYP1A expression was increased when the BaP concentration was 0.32mg/kg weight, but it was decreased with the increasing concentration of BaP. A similar tendency was observed in the preheated mice, but had higher CYP1A expression than in BaP group. The lung tissue of all mice treated with BaP had inflammatory changes. Conclusion The CYP1A protein was induced in lung tissue of mice treated with low concentration BaP, while high concentration of BaP could inhibited the expression of CYP1A.
