摘 要:目的探讨转化生长因子-β1(transforminggrowthfactor-β1,TGF-β1)在慢性硬化性颌下腺炎中的表达及其与颌下腺纤维化之间的关系。方法收集慢性硬化性颌下腺炎患者44例,根据颌下腺纤维化程度分期分为四组,用免疫组织化学方法分别观察四组中的TGF-β1、Ⅰ型胶原(collagenⅠ,COL-Ⅰ)和纤维连接蛋白(fibronectin,FN)的表达,并与正常组对照,用图像分析仪测定单位面积阳性颗粒百分比并进行统计学分析。结果TGF-β1、COL-Ⅰ和FN的表达量均与颌下腺的病变程度成正比(各组均P<0.05);此外TGF-β1与COL-Ⅰ、FN的表达量成正相关(各组均P<0.01)。结论随着慢性硬化性颌下腺炎病变程度的加重,TGF-β1表达显著增加,它可能通过促进细胞外基质(extracellularmatrix,ECM)主要成分如COL-Ⅰ、FN等合成的增加,而参与颌下腺纤维化发生,并在颌下腺纤维化形成过程中起重要作用。Objective To study the expression and significance of TGF-β1 in chronic sclerosing sialadenitis of the submandibular gland. Methods Expression of transforming growth factor-β1 (TGF -β1), collagen Ⅰ (COL - Ⅰ ), and fibronectin (FN) were investigated with immunohistochemistry assays in 44 patients with chronic sclerosing sialadenitis of submandibular gland. These patients were graded into four- degree groups according to the extent of fibrosis. Results The study found that the expression intensity of TGF-β1 ,COL- Ⅰ,and FN was correlated to the fibrotic grade of the subrnandibular gland ( P 〈0.05). The expression of TGF-β1 in subrnandibular injured area was correlated with that of the COL - Ⅰ( P 〈 0.01 ) and FN ( P 〈 0.01 ). Conclusion The increased expression of TGF-β1 may play an important role in the development and progression of the submandibular fibrosis by increasing the synthesis and accumulation of extracellular matrix (ECM).
