摘 要:目的:研究整合素β1(integrinβ1)、基质金属蛋白酶9(MMP9)、血管内皮生长因子(VEGF)在前列腺癌组织中的表达,探讨其与肿瘤侵袭转移的关系及相关性。方法:采用免疫组织化学染色(SP法)分别检测integrinβ1,MMP9,VEGF在15例良性前列腺增生及39例前列腺癌组织中的表达。结果:在良性前列腺增生和前列腺癌组织中integrinβ1蛋白的阳性表达率分别为80.00%和46.15%;MMP9蛋白的阳性表达率分别为33.33%和82.05%;VEGF蛋白的阳性表达率分别为46.67%和82.05%。3种蛋白在良性前列腺增生和前列腺癌组织中的阳性表达差异均有显著性(P<0.05,P<0.01)。3种蛋白的阳性表达均与肿瘤的临床分期有关(P<0.05,P<0.01),但都与前列腺癌的组织学分级无关(P>0.05)。in-tegrinβ1与VEGF之间的阳性表达呈负相关,MMP9与VEGF之间的阳性表达呈正相关。结论:本研究显示:integrinβ1,MMP9,VEGF可能共同参与了前列腺癌的浸润转移过程,联合检测integrinβ1,MMP9,VEGF的表达有利于更好的评估前列腺癌的生物学行为,可作为前列腺癌预后判断的有用指标。Objective: To investigate the expression of integrin β1, atrixmetalloproteinase ( MMP9 ), and vascular endothelial growth factor (VEGF) in Human Prostatic cancer and its relationship with invasion and metastasis of tumor, and to analyze the correlation among the expression of the 3 proteins. Methods : Immunohistochemical method was used to detect the expression of the 3 proteins in 15 cases of benign prostate hyperplasia and 39 cases of prostatic cancer. Results: In the benign prostate hyperplasia cases and prostatic cancer cases, the positive expression of integrin β1, MMP9 and VEGF were 80.00% and 46.15%, 33.33% and 82.05%, and 46.67% and 82.05% respectively. There were significant differences of the expression of the 3 proteins between benign prostate hyperplasia and prostatic cancer (P 〈0. 05, P 〈0. 01 ). The positive expression of all the tree correlated with clinical stages of the diseases (P 〈0. 05, P 〈0. 01 ), but no correlation with the pathological differentiation was found (P 〉 0.05 ). Negative correlation existed between expression of integrin β1 and that of VEGF, and a positive correlation existed between expression of MMP9 and that of VEGF in prostatic cancer tissue. Conclusions: The investigation results show that integrin β1, MMP9, and VEGF might be involved in the process of the infiltration and metastasis of prostatic cancer. The combined reference of expression of integrin β1, MMP9, and VEGF benefits the better inference on the biological behavior of of prostatic cancer and could be used as good indicators for the prognosis predication of the disease.
