转化生长因子-β_1及其受体表达与白血病病情的相关性研究

摘　　要：目的探讨急性白血病患者的转化生长因子-β1(TGF-β1)及其受体表达与白血病治疗、预后的相关性。方法收集42例初诊和经化疗完全缓解后白血病患者的外周血和骨髓,用ELISA法检测外周血、骨髓及骨髓基质细胞的可溶性TGF-β1含量;用免疫组织化学法检测外周血白血病细胞和骨髓有核细胞上的TGF-β1及其Ⅱ受体(TGFR2)。结果急性白血病患者外周血、骨髓、骨髓基质细胞的TGF-β1含量明显低于对照(P<0·01),尤以血清为甚,经化疗缓解后,TGF-β1含量又升高至接近正常水平。外周血白血病细胞和骨髓有核细胞上的TGF-β1表达与对照组比较无明显差异(P>0·05)。初诊患者外周血TGFR2表达明显低下,缓解后,TGFR2表达又升高至接近正常。结论急性白血病血浆、骨髓、骨髓基质TGF-β1含量与TGFR2与临床病情密切相关,可作为了解病情、观察疗效、判断预后的指标之一。Objective To study the relationships between transforming growth factor-β1 （TGF-β1）,TGFR2 and clinical nosography in different stage of leukemic patients. Methods Peripheral and bone marrow blood at first visit and complete recuperate of acute leukemic patients were collected. Soluble TGF-β1 was detected by enzymelinked immunoassay（ELISA） in peripheral blood,bone marrow blood and the supernatant of cultured bone marrow stromal cells,and membrane-binding type TGF β1, TGFR2 on the surface of peripheral blood luekocytes and bone marrow nucleared ceils were detected by immunohistocheminstry. Results Soluble TGF-β1 in peripheral blood,bond marrow blood and the supernatant of cultured bone marrow stromal cells decreased significantly compared with the contral group（P 〈 0.01 ）, especially in peripheral blood. The soluble TGF-β1 returned to nomal level after complete recuperate. The menmbrane-binding type TGF-β1 in peripheral and in bone marrow nucleated cells had got little change（ P 〉 0.05）. Differently, cell-binding TGFR2 on peripheral blood luekocytes decreased observably at first visit, it increased to normal level after complete recuperate. Conclusions There is an obvious relationship between the soluble TGF-β1, membraneobinding type TGFR2 in plasma of peripheral blood, bone marrow blood, the supernatant of bone marrow stromal cells and clinical of leukemic patients. TGF-β1 and TGFR2 maybe an valuable parameter to estimate the prognosis of leukemic patients clinically.
