摘 要:目的:探讨新生鼠缺氧缺血性脑损伤后早期不同时间点肾上腺髓质素基因表达的改变。方法:实验于2004-03/12在天津医科大学毒理实验室和天津市肿瘤医院中心实验室进行。①分组:取80只新生7dWistar大鼠,随机分为10组,对照组、缺氧缺血0,2,4,6,8,10,12,24,48h组,每组8只。②造模:对照组不结扎也不缺氧,其他各组大鼠结扎左颈总动脉2h后置于2500mL密闭玻璃容器中,以2.0L/min的速度输入体积分数为0.08的氧气2h制备大鼠缺氧缺血性脑损伤模型。③观察指标:应用反转录-聚合酶链反应方法测定各组大鼠脑皮质肾上腺髓质素mRNA的表达,以肾上腺髓质素mRNA/β-actin比值来表示。结果:经补充后80只大鼠进入结果分析。对照组肾上腺髓质素mRNA少量表达,缺氧缺血4h组明显高于对照组,缺氧缺血后8h表达达到最高峰(0.406±0.092,0.680±0.165,1.180±0.218,P<0.01),其后下降,缺氧缺血48h组与对照组无差异(0.518±0.153)。结论:新生大鼠发生缺氧缺血性脑损伤后肾上腺髓质素系统可能参与其后的病理生理过程,且肾上腺髓质素能作为早期判断缺氧缺血性脑损伤的一种诊断标志物。AIM: To investigate the expression of adrenomedulin (ADM) mRNA at different time points in newborn rats with hypoxic-ischemic brain damage (HIND). METHODS: The experiment was conducted in the laboratory for toxicology of Tianji Medical University and central laboratory of Tianjin Cancer Hospital from March to December 2004. ①Grouping: Eighteen neonatal 7-day-old Wistar rats were randomly divided into 10 groups: control group, 0-hour, 2-hour, 4-hour, 6-hour, 8-hour, 10-hour, 12-hour and 24-hour hypoxia-ischemia groups, 8 rats in each group. ②Modeling: All the rats were ligated on the left common carotid artery, and then placed in an obturating glass bottle(2 500 mL) 2 hours later followed by the input of 80 mL/L oxygen at 2.0 L per minute to induce HIBD except those in the control group.③ Observation indexes: The expression of ADM mRNA was measured in cortex at different time points by the reverse-transcriptionpolymerase chain reaction (RT-PCR), and expressed by the ratio of ADM mRNA to β-actin. RESULTS: Totally 80 rats were analyzed in the result after supplement. The expression of MDM mRNA was a little in the control group, increased in the 4-hour hypoxia-ischemia group, and peaked in the 8-hour hypoxia- ischemia group(0.406±0.092, 0.680±0.165, 1.180±0.218, P 〈 0.01). The expression of MDM mRNA was decreased gradually from the 84 hour after hypoxia-ischemia, and when at 48 hours, it was insignificantly different from that in the control group(0.518±0.153). CONCLUSION: ADM system not only involves in the occurrence of HInD in neonatal rats, but also may be a diagnostic indicator of early HIND.
