雌激素对沙土鼠脑缺血再灌注后脑海马CA1区细胞凋亡的影响

摘  要:目的:应用单因素设计观察雌激素对去卵巢沙土鼠脑缺血再灌注后脑海马CAI区细胞凋亡的影响。方法:实验于2004-10/2005-01在菏泽市立医院中心实验室完成,将45只雌性沙土鼠随机分为假手术组、卵巢切除组和雌二醇组3组,每组15只。①预处理:卵巢切除组和雌二醇在实验前2周切除双侧卵巢,假手术组手术但不切除卵巢。雌二醇组自切除卵巢次日起,每天腹腔注射雌二醇(0.1mg/kg),连续2周;其他两组每日腹腔注射0.5mL的生理盐水。②模型制备:3组均采用双侧颈动脉夹闭法制备脑缺血再灌注模型。③观察指标:所有动物造模后3d麻醉状态下处死取脑,测定脑组织匀浆中超氧化物歧化酶及丙二醛水平,TdT介导的原位末端标记法计量分析。结果:45只沙土鼠全部进入结果分析。①脑海马CA1区细胞凋亡密度:卵巢切除组高于假手术组和雌二醇组[(67.70±5.98),(43.46±4.66),(45.13±3.87)个/40×10视野,F=21.32,P<0.05]。②脑组织中丙二醛浓度:卵巢切除组高于假手术组和雌二醇组[(4.67±0.56),(2.74±0.25),(2.77±0.31)μmol/L,F=23.56,P<0.05]。③超氧化物歧化酶活性:卵巢切除组低于假手术组和雌二醇组[(38.12±4.17),(56.22±7.80),(54.78±6.90)NU/mL,F=19.75,P<0.05]。假手术组与雌二醇组比较上述指标均无差异(P>0.05)。结论:雌激素可以减少自由基损伤时脑神经元中的丙二醛含量,提高超氧化物歧化酶活性,减轻沙土鼠脑缺血再灌注后脑海马CA1区神经细胞的凋亡,对沙土鼠脑缺血再灌注损伤具有保护作用。AIM: To observe the effects of estrogen on the apoptosis in the CAI region of cerebral hippocampus after cerebral ischemia/reperfusion in ovariectomized female gerbils. METHODS: The experiment was performed in the Central Laboratory of Heze Municipal Hospital from October 2004 to January 2005. Forty-five female gerbils were divided into three groups at random: sham-operated group, ovariectomized group and aquadiol group with 15 in each group. ① Pretreatment: The ovary on the both side were cut at 2 weeks before experiment in the ovariectomized group and aquadiol group. Those in the sham-operated group were performed operation without cutting the ovary. The gerbils in the aquadiol group were treated with aquadiol (0.1 mg/kg) by intraperitoneal injection from the second day of cutting the ovary every day for 2 weeks. Those in other two grnups were treated with 0.5 mL saline through intraperitoneal injection every day.② Preparation of model: The gerbils in the three groups were treated with double side carotid closure method to make cerebral ischemia/reperfusion model. ③ Observation index: All the animals were killed under drugged state at 3 days after making models to gain the brain. The levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in the homngenate of brain tissue were detected. The in situ end-labeling (ISEL) mediated by TdT was conducted to calculate analysis. RESULTS: Totally 45 gerbils were involved in the result analysis. ① The density of apoptosis in the CA1 region of the cerebral hippoeampus: It was higher in the ovariectomized group than that in the sham-operated group and the aquadiol group [(67.70±5.98), (43.46±4.66), (45.13±3.87)per 40×10 visual field, F=21.32, P 〈 0.05].② Concentration of MDA in the brain tissue: It was higher in the ovariectomized group than that in the sham-operlted mgrop and the aquadiol group[ (4.67±0.56), (2.74±0.25), (2.77±0.3 )μmol/L,F=23.56 P 〈 0.05].③ Activity of SOD: It was l
