摘 要:目的:观察闭合性脑损伤后脑组织肿瘤坏死因子αmRNA表达变化,探讨对脑组织的影响。方法:实验于2004-09/2005-03在郑州大学基础医学院人体解剖学教研室神经分子生物学实验室进行,选择SD大鼠72只,随机分为实验组66只和对照组6只,实验组采用Marmarou法制备闭合性颅脑损伤模型,对照组不做任何处理。实验组又分为脑损伤后30min,1,3,9,12和1,2,3,4,5,6d11个时间点,每个时间点6只。应用反转录多聚酶链反应检测脑损伤后不同时间点脑组织中肿瘤坏死因子αmRNA表达。结果:72只大鼠全部进入结果分析。对照组未见肿瘤坏死因子α表达实验组损伤后30min及1h组与对照组无明显差别,9h~5d明显增加,12h~3d达到高峰,6d后开始下降。结论:大鼠闭合性脑损伤后脑组织肿瘤坏死因子αmRNA表达水平增加,是加重继发性脑损伤的重要因素。AIM: To observe the changes of the expression of tumor necrosis factor alpha (TNFα) mRNA in brain tissue after closed injury of brain, and explore its effect on brain tissue. METHODS: The experiment was conducted in the laboratory of Neural Molecular Biology of Staff Room of Human Anatomy of Basic Medical College of Zhengzhou University from September 2004 to March 2005. Seventy-two SD rats were selected and assigned randomly into experimental group with 66 rats and control group with 6 rats. The rats in experimental group were made into closed craniocerebral injury models with Marmarou method, The rats in control group were not treated with any disposal, The rats in experimental group were divided into 11 time points after brain injury, which were 30 minutes, 1, 3, 9, 12 hours and 1, 2, 3, 4, 5, 6 days with 6 rats in each time points. The expression of tumor necrosis factor alpha mRNA in brain tissue were detected after brain injury at different time point with reverse transcription polymerase chain reaction (RT-PCR). RESULTS: Totally 72 rats were involved in the result analysis, There were not the expressions of tumor necrosis factor alpha in control group. There was insignificant difference between 30 minutes and 1 hour group after injury in experimental group and control group, but it increased significantly from 9 hours to 5 days, and reached the peak from 12 hours to 3 days, and then began to decrease after 6 days. CONCLUSION: The increase of the expression levels of tumor necrosis factor α mRNA in brain tissue after closed injury of brain in rats is the important factor of adding the continuous brain injury.
