摘  要:本文对SD大鼠子宫内膜提取物作CRH放射免疫分析、用免疫组织化学原位定位技术研究CRH在大鼠子宫组织的分布,并采用自体对照的大鼠子宫角注射技术观察了CRH抗血清对胚泡着床的影响。结果证实:(1)妊娠和非妊娠大鼠子宫内膜都含有CRH样物质,其含量分别为:非妊娠大鼠3.63±0.32pg/mg湿组织(n=10),妊娠第4天大鼠4.57±0.52pg/mg湿组织(n=8),妊娠第7天大鼠4.38±0.49pg/mg湿组织(n=7)。妊娠大鼠与非妊娠大鼠间无显著差异。(2)CRH样免疫活性物质存在于子宫内膜固有层结缔组织的若干细胞和内膜腔面的柱状上皮细胞内。(3)当一侧子宫角注射10μlCRH抗血清,对侧子宫角注射10μl正常兔血清(NRS)时(n=6),可见抗血清侧胚泡着床率略高于NRS侧,但两侧相比差异不显著;而当一侧子宫角注射20μlCRH抗血清,对侧子宫角注射20μlNRS时(n=13),则抗血清侧胚泡着床率明显高于NRS侧(P<0.05)。一侧子宫角注射20μl生理盐水或NRS与假手术的对侧子宫角相比,着床率无明显差异。结果表明,CRH抗血清子宫角局部给药有提高胚泡着床率的作用,其作用方式可能是CRH抗血?xtracts of endometrium of Sprague-Dawley rats were measured by the method of radioim-munoassay for corticotropin- releasing hormone(CRH).The distribution of immunoreactive CRHwas observed bv immunohistochemistry technique and the effects of CRH- antiserum on blasto-cyst implantation were observed by local intrauterine administration.The results showed that:(1)CRH-like peptide exists in the endometrium of both pregnant and nonpregnant rats,(2)Im-munoreactive CRH was loCalized both in some cells of the proper lamina and columnar epitheliumin the endometrium.(3)When left uterine horn was inlected with CRH-antiserum and right uter-ine horn inlected with normal rabbits serum(NRS)on day 4 of pregnancy.in 10 μl group(n= 6).the implantation percentage in antiserum side was slightly higher than that in NRS side. but nosignificant difference appears between the both sides.In 20μlgroup(n=13).however.the im-plantation percentage in antiserum side was significantly higher than that in NRS side(P<0.05).(4)No significant change of implantation percentage was brought by injecting 20 μl ofNRS or normal saline when compared to sham-operation.These results suggested that CRH-likepeptide synthesized locally in the endometrium exerted inhibitory effects on blastocyst implanta-tlon.
