摘  要:运用PCR-SSCP-EB染色法对30例结直肠腺癌和18例结直肠腺瘤中P_53基因外显子5、6、7、8突变分别进行检测,结果显示腺癌、腺瘤中P_53基因突变率分别为66.7%和33.3%(P<0.05),高、中、低分化腺癌中P_53基因突变率分别为37.5%、62.5%和85.7%(P<0.05)。结果提示P_53基因突变与结直肠腺癌分化程度有关,在结直肠腺瘤向腺癌恶变过程中可能起着重要作用。PCR-SSCP-EB染色法是一种快速、简单且安全地检测基因突变的方法,适合在基层医院推广应用。Mutations of exon 5 to 8 of p53 gene were detected in 30 cases of colorectal adenocarcino- mas and 18 cases of colorectal adenomas by PCR-SSCP-EB (ethidium bromide) staining. Mutations of p53 gene occurred in 66.7% col- orectal adenocarcinomas and in 33.3% col- orectal adenomas (P<0.05). Mutation per- centages of p53 gene were 37.5%, 62.5% and 85% in well-differentiated adenocarcinomas respectively (P<0.05). It was suggested that- mutations of p53 were associated with the dif- ferentiation degrees of colorectal adenocarci- nomas and may play an important role in the progression of adenomas to adenocarcinomas. PCR-SSCP-EB staining was a quick, simple and safe way to detect gene mutations, it may find wide application in hospitals.
