摘 要:目的探讨反义NHE1基因转染对SGC-7901胃癌细胞形态学的影响及其在肿瘤基因治疗中的价值。方法构建人NHE1基因反义真核表达载体,采用脂质体法将其转染至SGC-7901胃癌细胞中,从光镜、电镜水平比较观察转染与未转染细胞形态学的变化。结果与亲本细胞相比,光镜下反义NHE1基因转染的SGC-7901细胞胞体增大,胞浆丰富,核大小相对一致,核浆比减少,核分裂相减少,异型性减小。透射电镜下细胞线粒体数量增多、体积增大,并可见髓鞘样变,粗面内质网、Golgi较发达,内质网扩张,核偏向一侧,极性增强。结论反义NHE1基因转染能使SGC-7901细胞发生形态学及超微结构的变化,有促进SGC-7901细胞分化的作用。Objective Io study the ettects ot the transetection ot antisense human NHE1 on the morphology of SGC-7901 and its potential role in the genetherapy for cancers, Methods Antisense NHE1 eukaryotic expression on vector pcDNA3.1 was constructed with recombinant DNA technique and transfected into SGC-7901 with liposome DOTAP. We examined the morphological changes of the transfected cells under light and electron microscopy, Results Under light microscopy , transfected cells with antisense gene to NHE1 were larger with abundant cytoplasm and nuclei division decreased . The ratio of nuclei to cytoplasm increased as compared with their parent cells. Under electron microscopy, organcelles in these cells were abundant. Conclusion Our study demonstrats that after the transfection of antisense NHE1 the morphology of SGC-7901 changes, NHE1 antisense gene cand promote differentiation of SGC-7901.
