汕优63的DNA分子标记及种子纯度鉴定研究

摘　　要：利用改进了的ＤＮＡ提取方法，从“三系”杂交水稻汕优６３的不育系、保持系、杂种一代和恢复系单株中提取ＤＮＡ，进行特异基因片断的ＰＣＲ基因扩增分析。结果表明：从２００个随机引物和设计的特定引物中筛选出３个引物，能有效地从不育系中鉴别出保持系，从汕优６３中分别鉴定出不育系、保持系、恢复系。研究结果表明，利用这些ＤＮＡ分子标记鉴定种子纯度，具有快速、准确、可靠的优点。The PCR analysis for the seed Durity detection of hybrid rice“Shanvou 63” isreportcd in this paper.By the use of motified CTAB method,the DNA samples from riccseedlings were extracted effectively.From 200 random primers and 7 special primers.three primers PFA,PFB,PAB,were selected for seed purity detection.By using PFAaid PFB,we can identify the male sterile line(Zhen-Shan 97A)and restore line(MingHui 63)seeds from hybrid seeds(Shan-You 63);and by using PAB we can distinguish themale sterile line and maintainer line seeds (Zhen-Shan 97B)from each otber.The resultsindicate that PCR analysis is an effective,accutive and simple way for seed purity detec-tion.
