摘 要:目的观察细菌脂多糖(lipopolysaccharide,LPS)、脂蛋白(bacteriallipoprotein,BLP)及其DNA〔鸟嘌呤二核苷酸(CpG)寡核苷酸(oligonucleoetide),CpG-ODN〕的体内协同致病作用。方法147只小鼠随机分为等渗盐水对照组、单纯LPS组、单纯CpG-ODN组、单纯BLP组、LPS+BLP组、LPS+CpG-ODN组和LPS+BLP+CpG-ODN组,尾静脉注射给药,观察小鼠死亡率及血清肿瘤坏死因子(TNF)-α水平。结果注射后12,24,48h,二联组的死亡率显著高于单独给药组(P<0.05或P<0.01);三联给药组各时相点死亡率显著高于二联组(P<0.05)。单独给药后6h,TNF-α水平均显著高于对照组(P<0.05),12,24h与对照组比较,差异无统计学意义。二联组与单因素组和对照组比较,6,12h血TNF-α均明显升高,差异有统计学意义(P<0.05)。三者联合刺激时,6,12h血TNF-α与二联组比较,差异有统计学意义(P<0.05),至24h仍显著高于对照组(P<0.05)。结论细菌LPS、细菌脂蛋白和细菌DNA不仅自身存在一定的致病能力,而且三者间具有明显的协同效应,特别是三者联合给药时作用最强。Objective To observe the synergistic pathopoiesis effects of lipopolysaccharide ( LPS), bacterial lipoprotein (BLP) and bacterial DNA in vivo. Methods A total of 147 mice were randomly divided into seven groups, ie, normal saline control group, simple LPS group, simple CpG-ODN group, simple BLP group, LPS plus BLP group, LPS plus CpG-ODN group and LPS plus BLP plus CpG- ODN group. LPS, BLP and bacterial DNA were injected via mice caudal vein to observe the mice mortality and the level of serum tumor necrosis factor-α (TNF-α). Results The mice mortality rates in the LPS plus BLP group and the LPS plus CpG-ODN group were significantly increased at the 12th, 24th and 48th hours respectively after injecting LPS, BLP and bacterial DNA, (P 〈 0.05 or P 〈 0.01 ) , when the mortality rate in the LPS plus BLP plus CpG-ODN group was much higher than that in the LPS + BLP group and the LPS plus CpG-ODN group (P 〈0.05). Compared with the control group, the serum TNF-α levels in groups of simple LPS, BLP and CpG-ODN were significantly elevated at the 6th hour ( P 〈 0.05 ) but decreased gradually near to normal level at the 12th and 24th hours. In the LPS plus BLP group and the LPS plus CpG-ODN group, the serum TNF-α level was much higher than that in simple injection groups and control group at the 6th and 12th hours, with significant difference (P 〈 0.05). The serum TNF-α levels in the LPS plus CpG-ODN plus BLP group was significantly increased at the 6th and 12th hours, with significant difference compared to the LPS plus BLP group and the LPS plus CpG-ODN group ( P 〈 0.05 ) , and still higher compared to the control group (P 〈 0.05 ). Conclusion LPS, BLP and bacterial DNA in itself not only has toxic insult to mice, but they together have obvious synergistic pathopoiesis effects.
