摘 要:目的:探讨灵芝的主要有效成分灵芝多糖肽对H2O2氧化应激损伤PC12细胞的保护作用,并以神经生长因子作为阳性对照。方法:实验于2004-01/06在第二军医大学神经科学研究所进行。以H2O2损伤PC12细胞为氧化应激损伤神经元的模型,将培养的PC12细胞随机分成4组:①正常对照组:PC12细胞在无血清的DMEM培养基中培养。②H2O2作用组:PC12细胞与终浓度为200μmol/L的H2O2作用4h。③灵芝多糖肽组:PC12细胞预先用终浓度为10mg/L的灵芝多糖肽预处理24h后,加入终浓度为200μmol/L的H2O2共同作用4h。④神经生长因子组:PC12细胞预先用终浓度为0.1mg/L的神经生长因子预处理24h后,加入终浓度为200μmol/L的H2O2共同作用4h。采用MTT法检测细胞的存活率以及Westernblotting检测半胱氨酸天冬氨酸蛋白酶3P20活性片段。结果:①细胞存活率:H2O2作用组显著低于正常对照组犤(38.6±7.1)%,(100±9.3)%,P<0.01犦;灵芝多糖肽组和神经生长因子组显著高于H2O2作用组犤(83.4±10.2)%,(75.9±7.4)%,P<0.01犦,但两组间比较无差异(P>0.05)。②半胱氨酸天冬氨酸蛋白酶3P20活性片段:Westernblotting检测结果提示灵芝多糖肽组和神经生长因子组较H2O2作用组明显减少。结论:适量灵芝多糖肽灵芝多糖肽能促进氧化应激损伤PC12细胞的存活,抑制H2O2诱导的PC12细胞凋亡,对氧化应激损伤PC12细胞具有保护作用,其效果与神经生长因子相似。AIM: To investigate the protective effects of ganoderma lucidum polysaccharides peptide, which is the effective component of ganoderma, on damaged PC12 cells induced by H2O2, oxidative stress, and take nerve growth factor as positive control. METHODS: The experiment was carried out in the Institute of Neuroscience, the Second Military Medical Universily of Chinese PLA between January and June 2004. H2O2, induced damaged PC12 cells were used as the models of neuron damage induced hy oxidative stress, and the cuhured PC12 cells were randomized into four groups: ① normal controt group: the PC12 cells were cuhured in DMEM medium without serum;② H2O2 trealment group: the PC12 cells were treated with H2O2 (the terminal concentration was 200μmol/L) for 4 hours;③ ganoderma lucidum polysaccharides peptide treatment group: the PC12 cells were treated with ganoderma lucidum polysaccharides peptide (the terminal concentration was 10μg/mL) for 24 hours, and then H2O2 (the terminal concentration was 200μmol/L) was also added for 4 hours; ④ nerve growth factor treatment group: the PC12 cells were treated with ganoderma lueidum polysaecharides peptide (the terminal concentration was 0.1 mg/L) for 24 hours, and then H2O2, (the terminal coneentratiun was 200μmol/L) was also added for 4 hours. The survival rate of the PC12 cells was detected with MTT assay. The activity of Caspase-3 P20 was determined with Western blotting. RESULTS: ① The survival rate of PC12 cells: it was significantly lower in the H2O2 treatment group than in the normal cuntrul group [(38.6±7.1)%, (100±9.3)%, P 〈 0.01), siguifieantly higher in the ganoderma lucidum polysaeeharides peptide treatment group and nerve growth factor treatment group than in the H2O2treatment group [(83.4±10.2)%, (75.9±7.4)%, P 〈 0.01], and there was insignificant difference between the two groups (P 〉 0.05).②The activity of Caspase-3 P20: The results uf Western blotting showed that it was obviously
