控制性超排卵明显降低小鼠卵母细胞水通道蛋白的表达

摘　　要：目的了解控制性超排卵(COH)对小鼠卵母细胞上表达水通道蛋白(AQP3)的影响。方法模拟人类超排卵周期,对小鼠进行超促排卵,按随机表顺序分为COH组和对照组,每组各10只ICR小鼠,用半定量实时荧光PCR方法检测小鼠MⅡ卵母细胞上AQP3mRNA的表达。结果卵母细胞膨胀实验显示,ICR小鼠卵母细胞上有AQP3的表达,用半定量实时荧光PCR方法检测时以β-肌动蛋白(β-actin)作为内参照,COH组和对照组ΔCT值分别是0·65±0·23和2·49±0·05,ΔΔCT值是1·84±0·18,两组比较P<0·001,具有统计学意义。结论控制性超排卵(COH)明显降低了ICR小鼠卵母细胞上AQP3mRNA的表达,这也可能是导致卵母细胞低冻融复苏率低的原因之一。Objective To study the effect of controlled ovarian Hyperstimulation（COH）on expression of AQP3 mRNA in mouse oocytes at metaphase Ⅱ . Methods Twenty female mice（6-7 weeks） were randomly allocated into 2 groups, mice in COH group were superovulated by intraperitoneal injection of 7.5 IU pregnant mare＇ s serum gonadotropin （PMSG） followed by 5 IU human chorionic gonadotropin（HCG） after 46-48 h. Nothing was given to mice in control group and the estrus cycle was observed at 9 am everyday. 12-16 h following hCG injection （COH group） or at 8 am next day after the estrus （control group）, mice were killed by cervical dislocation. The oviducts were excised. Cumulus masses were recovered from the dilated ampullae under a dissecting microscope,digested granulosa cells using hyaluronidase. Semi-quantitative real-time PCR of AQP3 mRNA in mouse M Ⅱ oocytes was investigated with β-aetin as the internal control. Results Oocytes swelling assay showed that AQP3 mRNA expressed in mouse M Ⅱ oocytes. Using semi-quantitative real-time PCR, the expression of AQP3 mRNA was significandy decreased （ P 〈 0.001 ） in COH group（ ACT = 0.65 ± 0.23 ） compared with the control group （ACT = 2.49 ± 0.05 ）. Conclusion Controlled ovarian hyperstimulation significandy decreased the expression of AQP3 mRNA in mouse M Ⅱ oocytes. May be this was one of reasons of the low survival of oocytes after cryopreservation.
